Depends.
IMO, Guitar Hero allows you to be a little sloppier and be successful. Rock Band you pretty much have to be dead on.
We have GH for the Wii and RB for the PS3...
you also have to look at compatibility and future compatibility. Guitar hero seems to let you play with all the games and guitars and all that. also, check the songs you want to play, sing

Visually, I prefer GH.
RB is nice in that you can have two people playing guitar in the 'band'.
GH, one would have to play bass.
But I'm not that hip on either one, perhaps I've missed the way to do it in GH.

Either way, you'll need loads of booze. I prefer GH, but the gf prefers RB because she likes to sing. Neither of us nor any of her little friends like playing drums.
I prefer Rock Band... I guess I am the only one so far. I like the buttons on the RB guitar a lot better. Plus I have small hands. The RB guitar has 2 sets of buttons, one set near the top and one set near the base with buttons closer together.

We have been playing RB here on vacation in DC. I started off on drums and sucked!
I always thought I could not sing. I can on RB. Even on the medium setting I get high 90's. Even on Rush Tom Sawyer! I guess all those years of singing in the bubble bath are paying off.
Heck, we do not even have a TV at home. Getting a playstation would be a big leap for us.

I haven't A:Bed these games extensively, but I like the Guitar Hero guitars a little more since they give better feedback on your "strumming" - the Rock Band guitar goes "squish", the Guitar Hero guitar goes "clack". Also the tremolo (whammy bar) in the RB game is largely cosmetic, whereas in GH you get points for getting your whammy on. Going into bonus point mode in the game is a little easier to activate in GH (tipping your guitar up) compared to RB (pushing a button - a little harder than it sounds).
If you have ever played drums, you may like GH a little more - the layout (http://ecx.images-amazon.com/images/I/416Lo3vIMlL._SS400_.jpg) is more like a drum kit, with cymbals on top, and snare and toms on the bottom. There are third party kits you can buy if it really makes a difference to you.
I have RB, and I am glad I went that direction since they just released the Beatles game - GH has Metallica, on the other hand...
